New monoclonal antibody for the sensitive detection of hydroxy-s-triazines in water by enzyme-linked immunosorbent assay

Resumé

This article presents a new and sensitive immunoassay for the triazine metabolites hydroxypropazine, hydroxyatrazine and hydroxysimazine. The assay is based on covalent immobilisation of antigen in combination with an enzyme-labelled anti-hydroxy-s-triazine monoclonal antibody. This system enabled the development of an assay with variation coefficients below 3% and a detection limit below 0.01μg/l hydroxyatrazine and hydroxypropazine. Analysis of hydroxyatrazine-spiked water of three different types yielded an average recovery of 102% at 0.1μg/l hydroxyatrazine. Relative to hydroxyatrazine, assay cross-reactivity was 148% towards hydroxypropazine and 67% towards hydroxysimazine. Copyright (C) 2000 Elsevier Science B.V.
